Description

The 20S proteasome serves as the primary enzyme complex responsible for breaking down proteins within cells. To aid in its activity, three prominent protein complexes known as PA700, PA28, and PA200 act as activators for the 20S proteasome. Among these, PA28 consists of three evolutionarily conserved subunits: PA28α (PSME1), PA28β (PSME2), and PA28γ (PSME3) (1,2). PA28α and PA28β combine to form a heteroheptameric complex that facilitates binding to the opening site(s) of the 20S complex. This PA28α/β partnership is found throughout the cell and plays a crucial role in MHC class I antigen presentation by aiding in the creation of antigenic peptides derived from foreign proteins (2). On the other hand, PA28γ exists as a homoheptamer and primarily resides in the nucleus. The PA28γ complex functions by linking with specific nuclear target proteins and directing them towards the 20S proteasome for subsequent degradation (3,4).

Subunit of the 11S REG-gamma (also called PA28-gamma) proteasome regulator, a doughnut-shaped homoheptamer which associates with the proteasome. 11S REG-gamma activates the trypsin-like catalytic subunit of the proteasome but inhibits the chymotrypsin-like and postglutamyl-preferring (PGPH) subunits. Facilitates the MDM2-p53/TP53 interaction which promotes ubiquitination- and MDM2-dependent proteasomal degradation of p53/TP53, limiting its accumulation and resulting in inhibited apoptosis after DNA damage. May also be involved in cell cycle regulation. Mediates CCAR2 and CHEK2-dependent SIRT1 inhibition (PubMed:25361978).
