Background

Notch1 serves as a receptor for Jagged1, Jagged2, and Delta1 membrane-bound ligands, regulating cell-fate determination. Upon ligand activation, the released notch intracellular domain (NICD) forms a transcriptional activator complex with RBPJ/RBPSUH, initiating genes of the enhancer of split locus. Its functions include influencing differentiation, proliferation, and apoptotic programs and participating in angiogenesis by negatively regulating endothelial cell proliferation, migration, and angiogenic sprouting. Notch1 plays a critical role in CD4+ and CD8+ cell maturation, follicular differentiation, cerebellar development, and postimplantation development. Additionally, it may be involved in mesoderm development, somite formation, and neurogenesis and serve to enhance HIF1A function by sequestering HIF1AN away from HIF1A. Required for the THBS4 function in regulating protective astrogenesis from subventricular zone (SVZ) niche after injury, Notch1 is also integral for the determination of left/right symmetry by modulating the balance between motile and immotile (sensory) cilia at the left-right organiser (LRO).
