SDF-1 is a chemoattractant that specifically affects certain immune cells such as T-lymphocytes and monocytes, but not neutrophils. It activates a receptor called CXCR4, which leads to a rapid increase in calcium ions within the cells and promotes their movement towards the source of the chemoattractant. Additionally, SDF-1 binds to a unique receptor known as ACKR3, triggering a different signaling pathway called the beta-arrestin pathway. ACKR3 acts as both a scavenger receptor for SDF-1 and an activator of the beta-arrestin pathway. When SDF-1 binds to cell surface proteoglycans, it prevents the formation of certain SDF-1 isoforms, specifically SDF-1-alpha(3-67), thereby preserving its chemotactic activity in local areas. SDF-1 also serves as a positive regulator of monocyte migration and a negative regulator of monocyte adhesion through its interaction with the LYN kinase. By stimulating the migration of monocytes and T-lymphocytes via CXCR4 and ACKR3, SDF-1 reduces monocyte adherence to surfaces coated with ICAM-1, an integrin ligand. It achieves this by inhibiting the LFA-1 mediated adhesion of monocytes, a process mediated by the LYN kinase. Furthermore, SDF-1 inhibits the infection of T-cell line-adapted HIV-1 through its interaction with CXCR4. In the context of myocardial infarction, SDF-1 plays a protective role. It also plays critical roles during embryonic development, including facilitating B-cell lymphopoiesis, myelopoiesis in the bone marrow, and the formation of the ventricular septum in the heart.
