Overview

Description

Bax is a protein that plays a crucial role in the process of programmed cell death. It achieves this function by binding to certain proteins and impeding their activity. One such protein is BCL2, which is a potent inhibitor of apoptosis. Bax neutralizes the effect of BCL2, thereby enabling the cell to initiate the apoptotic process. It does this by prompting the release of cytochrome c and activating a protein called CASP3, both of which are critical components in the apoptotic pathway.
Bax belongs to the Bcl-2 family of proteins and exists primarily as a homodimer. However, it can also form heterodimers with other proteins in this family, such as BCL2L1 isoform Bcl-X(L), MCL1, and A1. Bax also interacts with various other proteins, such as SH3GLB1, HN, SFN, and YWHAZ. The latter two proteins bind to Bax in the cytoplasm and under stressful conditions, their phosphorylation via JNK triggers the release of Bax to mitochondria.
Interestingly, there are several different isoforms of Bax produced by alternative splicing. Isoform Sigma, for example, interacts with BCL2A1 and BCL2L1 isoform Bcl-X(L). Despite these variations in structure, all forms of Bax retain their essential function in promoting apoptosis.
