Overview

Description

Bim is a member of the BCL-2 protein family that serves as a pro-apoptotic factor. Its main function is to promote apoptosis or programmed cell death by interacting with other BCL-2 family members, such as BCL2, BCL2L1/BCL-X(L), and MCL1. Bim's expression can be induced by nerve growth factor (NGF) and the forkhead transcription factor FKHR-L1, indicating its involvement in neuronal and lymphocyte apoptosis.
In mouse studies, Bim was shown to be crucial in the process of thymocyte-negative selection, where it acts as an essential initiator of apoptosis. This underscores the significance of Bim in cellular regulation and highlights its potential as a therapeutic target for diseases associated with uncontrolled cell growth.
Overall, Bim's eight alternatively spliced transcript variants have been reported, reflecting its diverse biological roles and regulatory mechanisms. The intricate nature of Bim's function and regulation underscores the importance of deepening our understanding of Bim in normal and pathological conditions.

Other names

BCL2 like 11, also known as B2L11_HUMAN or BAM, is a protein that interacts with Bcl-2 and plays a crucial role in apoptosis. It is also referred to as Bcl 2 interacting protein Bim or Bcl 2 related ovarian death agonist. BCL2 like 11 is a facilitator of apoptosis and functions as a mediator of cell death. It is encoded by the BCL2L11 gene and is often abbreviated as BCL2L11 or BIM. Other names for this protein include Bcl2-interacting mediator of cell death, Bcl2-L-11, Bcl2l11, BIM alpha6, BIM beta6, BIM beta7, BimEL, BimL, and BOD.

Specificity

The BIM antibody has the ability to recognize and bind to the natural levels of total BIM present within the system. This property is crucial for accurate detection and analysis of BIM molecules in biological samples.

Stability & Storage

To ensure preservation, please store at a temperature of -20 °C for a duration of one year. It is important to avoid subjecting the product to multiple freeze/thaw cycles.

Storage buffer

The storage condition for Rabbit IgG is as follows: it should be kept in phosphate buffered saline with a pH of 7.4, containing 150mM NaCl, 0.02% sodium azide, and 50% glycerol. The recommended storage temperature is -20 °C. This antibody is stable for a period of 12 months from the date of receipt.

Target

Background

Induces apoptosis and anoikis. Isoform BimL is more potent than isoform BimEL. Isoform Bim-alpha1, isoform Bim-alpha2 and isoform Bim-alpha3 induce apoptosis, although less potent than isoform BimEL, isoform BimL and isoform BimS. Isoform Bim-gamma induces apoptosis. Isoform Bim-alpha3 induces apoptosis possibly through a caspase-mediated pathway. Isoform BimAC and isoform BimABC lack the ability to induce apoptosis.

Tissue specificity

Isoform BimEL, isoform BimL and isoform BimS are the predominant isoforms and are widely expressed with tissue-specific variation. Isoform Bim-gamma is most abundantly expressed in small intestine and colon, and in lower levels in spleen, prostate, testis, heart, liver and kidney.

Posttranslational modification

Phosphorylation at Ser-69 by MAPK1/MAPK3 leads to interaction with TRIM2 and polyubiquitination, followed by proteasomal degradation (PubMed:15486195, PubMed:21478148). Deubiquitination catalyzed by USP27X stabilizes the protein (By similarity).Ubiquitination by TRIM2 following phosphorylation by MAPK1/MAPK3 leads to proteasomal degradation. Conversely, deubiquitination catalyzed by USP27X stabilizes the protein.
