Description

Casp10 is a crucial protein involved in the mechanism of apoptosis execution mediated by caspases. Its activation cascade is triggered by both Fas and TNFR-1 receptors, which recruit Casp10 in a FADD dependent manner. In addition, it may also participate in the granzyme B apoptotic pathways. Casp10 is known to cleave and activate various caspases such as caspase-3, -4, -6, -7, -8, and -9. Furthermore, it is able to hydrolyze small-molecule substrates like Tyr-Val-Ala-Asp-|-AMC and Asp-Glu-Val-Asp-|-AMC.
Casp10 belongs to the peptidase C14A family and forms a heterotetramer consisting of two antiparallel arranged heterodimers. Each heterodimer is made up of a 23/17 kDa (p23/17) subunit (depending upon splicing events) and a 12 kDa (p12) subunit. Casp10 also has self-associating properties and interacts with FADD and CASP8. It is found in a Fas signaling complex that comprises of FAS, FADD, CASP8, and CASP10.
It is worth noting that human Casp10 has 6 isoforms that are produced through alternative splicing.
