Background

The gene in question encodes a versatile nuclear phosphoprotein with multifunctional roles in apoptosis, cell cycle progression, and cellular transformation. Specifically, it acts as a transcription factor that regulates the transcription of specific target genes. Various mutations, rearrangements, overexpressions, and translocations of this gene have been associated with hematopoietic tumors, leukemias, lymphomas, and Burkitt lymphoma. Interestingly, there are two isoforms of this protein resulting from alternative translation initiations from an upstream, in-frame non-AUG (CUG) and a downstream AUG start site. The N-termini of these isoforms are distinct, and evidence suggests that the synthesis of the non-AUG initiated protein is repressed in Burkitt's lymphomas, indicating its significance in the gene's normal function.
