Overview

Description

This is a description of a Peptide Affinity Purified Rabbit Polyclonal Antibody (Pab), meaning it is an antibody that has been produced from rabbit cells and has high specificity for a particular peptide. The antibody has been purified using affinity chromatography, a process that takes advantage of the binding strength between the antibody and the peptide to isolate and purify the antibody. This Pab can be used in various applications, such as immunohistochemistry, western blotting, and ELISA, to detect and quantify the target peptide in biological samples. By generating a highly similar content to the original text, the context and information of the original message can be conveyed to the reader in different words and a different format.

Other names

Nestin, NES;NES

Source

Rabbit

Specificity

The antibody targeting Nestin is derived from rabbits that were immunized with a synthetic peptide consisting of 1389-1416 amino acids from human Nestin, conjugated with KLH. To produce content that closely matches the original text, it is important to rephrase the information while still retaining the key points. Therefore, the Nestin antibody was developed utilizing a synthetic peptide conjugated with KLH, composed of 1389-1416 amino acids from the human Nestin protein. Rabbits were immunized with this peptide to produce the antibody targeting Nestin.

Stability & Storage

To ensure proper storage, refrigerate the item at a temperature between 2-8°C for a maximum of 2 weeks. If you need to store it for a longer period, it is recommended to keep it at -20°C in small aliquots in order to prevent the occurrence of freeze-thaw cycles.

Storage buffer

The antibody provided is a purified polyclonal antibody that is supplied in phosphate-buffered saline (PBS) containing 0.09% (W/V) sodium azide. It has undergone purification using a protein A column and further purification through peptide affinity purification.

Research area

Signal Transduction plays a crucial role in various fields of biology, including Cancer, Cardiovascular health, Developmental Biology, and Neuroscience. Understanding the mechanisms of signal transduction can provide valuable insights into the pathogenesis of diseases and the development of potential therapeutic strategies.
In the context of Cancer, signal transduction pathways are often dysregulated, leading to uncontrolled cell growth and tumor formation. By studying the intricate web of signaling molecules and their interactions, researchers aim to identify key targets for preventing or treating cancer.
Cardiovascular diseases, such as heart attacks and strokes, are also influenced by signal transduction processes. Abnormal signaling events can disrupt the delicate balance of blood vessel constriction and dilation, promoting the development of atherosclerosis and hypertension. Discovering the underlying signaling cascades can lead to the development of novel drugs targeting cardiovascular diseases.
Developmental Biology investigates how organisms grow and develop from a single cell to a complex, multicellular organism. Signal transduction pathways guide various cellular processes during embryonic development, including cell differentiation and tissue patterning. Studying these intricate signaling networks sheds light on the fundamental principles of life.
In the field of Neuroscience, signal transduction is crucial for communication between neurons and the functioning of the nervous system. Signal transduction processes allow neurons to transmit electrical and chemical signals, enabling sensory perception, motor control, and cognitive functions. Understanding the mechanisms that govern neuronal signaling helps researchers unravel the complexities of brain function and provides insights into neurological disorders.
In summary, signal transduction is a fundamental process that impacts diverse areas of biology, including Cancer, Cardiovascular health, Developmental Biology, and Neuroscience. Deeper insights into the intricate signaling pathways can lead to breakthroughs in disease prevention, diagnosis, and treatment, ultimately improving human health and well-being.

Target

Background

Promoting brain and eye development, this essential nutrient is responsible for dismantling phosphorylated vimentin intermediate filaments (IF) when cells divide. It also plays a crucial role in directing IF proteins and other cellular factors to the offspring cells during progenitor cell division. Moreover, it is indispensable for the survival, rejuvenation, and stimulated growth of neural progenitor cells (similarly).

Overlay histogram showing SH-SY5Y cells stained with abs104234 (green line). The cells were fixed with 2% paraformaldehyde (10 min) and then permeabilized with 90% methanol for 10 min. The cells were then icubated in 2% bovine serum albumin to block non-specific protein-protein interactions followed by the antibody (abs104234, 1:25 dilution) for 60 min at 37ºC. The secondary antibody used was Goat-Anti-Rabbit IgG, DyLight® 488 Conjugated Highly Cross-Adsorbed(NA168821) at 1/400 dilution for 40 min at 37ºC. Isotype control antibody (blue line) was rabbit IgG (1μg/1x10^6 cells) used under the same conditions. Acquisition of >10, 000 events was performed.

Abs104234 staining Nestin in Human heart tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with formaldehyde and blocked with 3% BSA for 0. 5 hour at room temperature; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody (1/25) for 1 hours at 37°C. A undiluted biotinylated goat polyvalent antibody was used as the secondary antibody.
