Description | Purified Rabbit Polyclonal Antibody (Pab) |
Other names | LGR5, GPR49, and GPR67 are all members of the leucine-rich repeat-containing G-protein coupled receptor family. These receptors, also known as GPCRs, play important roles in various cellular processes. LGR5, which is also referred to as GPR49, is a well-studied receptor involved in stem cell regulation and tissue maintenance. On the other hand, GPR49 and GPR67 are less explored but have been linked to cancer development and progression. The functions of GPR49 and GPR67 are still being investigated, and their specific roles in cellular signaling pathways are not yet fully understood. However, their presence in the GPCR family suggests their potential significance in cellular communication and response to external stimuli. The identification of these receptors, such as GPR49 and GPR67, highlights the complexity of GPCR signaling and the importance of further research in this field. As we continue to unravel the functions of these receptors, we may gain valuable insights that could lead to the development of novel therapeutic approaches for various diseases. |

Specificity | The LGR5 antibody described here is produced by immunizing rabbits with a synthetic peptide derived from human LGR5 protein. The peptide is conjugated with KLH (keyhole limpet hemocyanin) to enhance the immune response. The specific region of LGR5 used for immunization spans amino acids 689 to 719. This methodology ensures the production of an antibody that recognizes and binds to LGR5 with high specificity and affinity. |

Stability & Storage | To ensure optimal storage of this product, it should be kept refrigerated at a temperature between 2-8°C. This will help to maintain its potency and stability for up to two weeks. For longer-term storage, it is recommended to store the product in small aliquots at a temperature of -20°C. This will prevent the product from undergoing freeze-thaw cycles, which can potentially damage its effectiveness. By taking these storage precautions, you can help to ensure that your product remains usable and effective for as long as possible. |
Storage buffer | The purified polyclonal antibody is provided in PBS containing 0.09% (W/V) sodium azide. It is prepared using the Saturated Ammonium Sulfate (SAS) precipitation method, followed by dialysis against PBS. |

Target | |
Background | This receptor, found in both the intestinal epithelium and hair follicles, plays a crucial role in activating the canonical Wnt signaling pathway by binding to R-spondins. Upon binding, it interacts with phosphorylated LRP6 and frizzled receptors, triggering the expression of target genes and increasing Wnt signaling. Unlike typical G-protein coupled receptors, this receptor does not use heterotrimeric G-proteins to transduce signals. It is involved in the maintenance and development of adult intestinal stem cells during postembryonic development. |
Celluar localization | Cell membrane; Multi-pass membrane protein. Golgi apparatus, trans-Golgi network membrane; Multi-pass membrane protein. Note=Rapidly and constitutively internalized to the trans-Golgi network at steady state. Internalization to the trans- Golgi network may be the result of phosphorylation at Ser-861 and Ser-864; however, the phosphorylation event has not been proven (PubMed:23439653). |

Overlay histogram showing SH-SY5Y cells stained with abs106134 (green line). The cells were fixed with 2% paraformaldehyde (10 min). The cells were then icubated in 2% bovine serum albumin to block non-specific protein-protein interactions followed by the antibody (abs106134, 1:25 dilution) for 60 min at 37ºC. The secondary antibody used was Goat-Anti-Rabbit IgG, DyLight® 488 Conjugated Highly Cross-Adsorbed(NA168821) at 1/400 dilution for 40 min at 37ºC. Isotype control antibody (blue line) was rabbit IgG (1μg/1x10^6 cells) used under the same conditions. Acquisition of >10, 000 events was performed.

Fluorescent confocal image of HeLa cells stained with LGR5 (loop2) antibody. HeLa cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.2%, 30 min). Cells were then incubated with abs106134 LGR5 (loop2) primary antibody (1:100, 2 h at room temperature). For secondary antibody, Alexa Fluor® 488 conjugated donkey anti-rabbit antibody (green) was used (1:1000, 1h). Nuclei were counterstained with Hoechst 33342 (blue) (10 μg/ml, 5 min). Note the highly specific localization of the LGR5 immunosignal to the nucleus and cytoplasm, supported by Human Protein Atlas Data (http://www.proteinatlas.org/ENSG00000139292).
