Description

Killer cell immunoglobulin-like receptors (KIRs) are expressed by natural killer cells and subsets of T cells. These receptors are transmembrane glycoproteins. They are located on chromosome 19q13.4 within the leukocyte receptor complex (LRC), which is approximately 1 Mb in size. The KIR genes are polymorphic and highly homologous, meaning they have multiple variations and are very similar to each other. While the specific gene content of the KIR gene cluster can vary among different haplotypes, certain framework genes are present in all haplotypes, including KIR3DL3, KIR3DP1, KIR3DL4, and KIR3DL2.
The KIR proteins can be classified based on the number of extracellular immunoglobulin domains they possess, which can be either 2D or 3D. Additionally, they can be grouped based on whether they have a long (L) or short (S) cytoplasmic domain. KIR proteins with a long cytoplasmic domain transmit inhibitory signals when they bind to their ligands, using an immune tyrosine-based inhibitory motif (ITIM). On the other hand, KIR proteins with a short cytoplasmic domain do not have the ITIM motif but instead associate with the TYRO protein tyrosine kinase binding protein to transmit activating signals.
Several KIR proteins have been identified to have specific ligands within the subset of HLA class I molecules. This suggests that KIR proteins play an important role in regulating the immune response. The interaction between KIRs and their ligands is believed to be crucial in determining the activation or inhibition of natural killer cells and T cells, thus modulating immune responses.
