Testing Principle：

This ELISA kit is designed for the detection of CXCL13 using a double antibody sandwich method. The high affinity conjugate plate is pre-coated with a specific anti-human CXCL13 capture antibody. The process involves adding the standard substance, the sample to be tested, and a biotin-labeled detection antibody to the plate in succession. After thorough mixing and oscillation, the sample is incubated at room temperature for 2 hours. During this incubation period, CXCL13 in the sample binds to both the solid phase antibody and the detection antibody. The plate is then washed thoroughly to remove any unbound materials. Next, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is added to the plate. After another round of washing, a TMB color substrate is added and the plate is incubated in the dark at room temperature to initiate color development. The depth of the color reaction is directly proportional to the concentration of CXCL13 in the sample. Finally, a termination solution is added to stop the reaction, and the absorbance of the plate is measured at a detection wavelength of 450 nm (with a correction wavelength of 570-630 nm) using a microplate analyzer.
