Testing Principle:
The double antibody sandwich ELISA method is utilized in this kit. The conjugate plate already contains the specific anti-human CXCL13 Capture Antibody with high affinity. Firstly, the standard substance, the sample to be tested, and the biotin labeled detection antibody are sequentially added into the HRP-plate well. The mixture is thoroughly oscillated and mixed to ensure proper interaction between the CXCL13 in the sample and the solid phase antibody as well as the detection antibody. Subsequently, the sample is incubated at room temperature for a duration of 2 hours. Following the incubation period, the mixture is thoroughly washed to eliminate any unbound ingredients. To detect the presence of the CXCL13, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is introduced into the mixture. Another round of washing is performed before adding the TMB color substrate. The mixture is then incubated in a dark environment at room temperature, allowing the color to develop. The intensity of the color reaction is directly proportional to the concentration of CXCL13 present in the sample. To stop the reaction, a termination solution is added. Finally, the absorbance value is measured at a detection wavelength of 450 nm (with a correction wavelength of 570-630 nm) using a microplate analyzer.
