Testing Principle：

This particular kit utilizes the double antibody sandwich Enzyme-Linked Immunosorbent Assay (ELISA) technique. First, a specific anti-human CXCL13 Capture Antibody is provided on a highly-affinity conjugate plate. Then, the standard substance, along with the sample being tested and biotin-labeled detection antibody, are successively added onto the HRP-plate well. After shaking and mixing well, the sample is incubated at room temperature for two hours, allowing the CXCL13 in the sample to combine with the solid-phase antibody and detection antibody. Once thoroughly washed to remove all unbound substances, horseradish peroxidase-labeled streptavidin-HRP (SA-HRP) is included in the mixture. After once again washing well, TMB color substrate is added and allowed to incubate in a dark room at room temperature. The depth of color produced is directly proportional to the concentration of CXCL13 found in the sample. Finally, the reaction is terminated using a termination solution, and then the absorbance value is evaluated using a microplate analyzer set to 450 nm detection wavelength (with the correction wavelength set between 570-630 nm).
