Testing Principle：

The detection of CXCL13 in human samples is facilitated by using a double antibody sandwich ELISA kit. Firstly, a high affinity conjugate plate with prepackaged specific anti-human CXCL13 Capture Antibody is employed. The standard substance and the sample to be tested are then added successively, followed by the biotin labeled detection antibody, into the HRP-plate well. The mixture is fully oscillated and subsequently CXCL13 in the sample undergoes a 2-hour incubation process. During this time, the CXCL13 in the sample combines with the solid phase antibody and detection antibody. After thorough washing to eliminate any free and unbound substances, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is added. The mixture is then washed again and TMB color substrate is added. The mixture is then kept in the dark at room temperature for color development. The strength of the color reaction is directly proportional to the concentration of CXCL13 in the sample. The process is finished by adding the termination solution to stop the reaction. The absorbance value is measured using a microplate analyzer at 450 nm detection wavelength and correction wavelength 570-630 nm.

Background：

IL-17A or IL-17 is an important cytokine that belongs to the IL-17 family of cytokines. It is produced by helper T cells and can be triggered by IL-23. IL-17 cytokines play a vital role in recruiting monocytes and neutrophils to inflammatory sites by producing chemokines in tissues, thereby hindering the onset of inflammation. In response to an extracellular pathogen's invasion, IL-17, a pro-inflammatory cytokine, acts aggressively by destroying the intruder cells. Along with tumor necrosis factor and IL-1, IL-17 has synergistic effects that strengthen the immune system's defense mechanisms. Several autoimmune and immune-related diseases, including rheumatoid arthritis, asthma, lupus, allograft rejection, anti-tumor immunity, psoriasis, and multiple sclerosis, are linked to the IL-17 family.
