Testing Principle：

The double antibody sandwich ELISA kit utilizes a high affinity conjugate plate preloaded with specific anti-human CXCL13 Capture Antibody. To perform the assay, the standard substance, sample, and biotin-labeled detection antibody are sequentially added to the HRP-plate well. Following thorough oscillation and mixing, the sample is incubated for 2 hours at room temperature, allowing the CXCL13 in the sample to bind to both the solid phase antibody and the detection antibody. After a thorough washing step to remove any unbound components, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is added to the well. Another washing step follows, and then the TMB color substrate is introduced for color development. The intensity of the color reaction is directly proportional to the concentration of CXCL13 in the sample. To stop the reaction, a termination solution is added, and the absorbance value is measured at a detection wavelength of 450 nm (with a correction wavelength of 570-630 nm) using a microplate analyzer. This assay format enables accurate and sensitive detection of CXCL13 levels in the sample.

Background：

IgA, also known as Immunoglobulin A, is a crucial antibody involved in protecting mucosal immunity. It belongs to the alpha subtype of heavy chains. IgA exists in two subclasses, namely IgA1 and IgA2, and forms a dimer known as secretory IgA (sIgA). In terms of importance, sIgA outweighs other immunoglobulins in mucosal secretions while being less prevalent in blood. The main function of the secretory component within sIgA is to safeguard the immunoglobulin from degradation caused by proteolytic enzymes. Consequently, sIgA can thrive in the challenging gastrointestinal environment and effectively hinder microbial reproduction in bodily secretions. Additionally, it has the ability to reduce the inflammatory effects caused by other immunoglobulins. Nonetheless, IgA only exhibits weak activation and regulation capabilities over the complement system.
