Testing Principle

This assay kit uses a sandwich enzyme immunoassay technique to measure quantitative levels of human GM-CSF. The high affinity plate is pre-coated with an anti-human GM-CSF antibody. Test samples, standards, and biotin-labeled detection antibodies are added to the wells of the plate. After incubation, the GM-CSF present in the samples binds to the solid-phase antibodies and detection antibodies, forming immune complexes. The unbound materials are then washed away, and horseradish peroxidase-labeled streptavidin is added. After washing, a chromogenic substrate is introduced, which produces a color reaction in the dark. The reaction is halted with a stop solution, and the absorbance is measured at a wavelength of 450 nm with a reference correction wavelength of 540 nm or 570 nm.

Background:GM-CSF

GM-CSF, also known as CSF-2 or granulocyte-macrophage colony stimulating factor, belongs to the common β chain cytokine family. It is a 30 kDa polypeptide that exhibits polytrophic properties. Alongside IL-3 and IL-5, GM-CSF utilizes an α-helix conformation and contains two disulfide bonds within its structure. This cytokine is synthesized and released by activated immune cells, mesenchymal cells, and epithelial cells. It circulates within the body as variable glycosylated monomers.

GM-CSF is known to be upregulated in different cells during inflammation, such as encephalitis-causing T cells, allergen-exposed lung endothelial cells, and IgE-activated mast cells. The mature form of human GM-CSF shares about 54% and 63% of its amino acid sequence with mouse and rat GM-CSF, respectively. The high-affinity receptor of GM-CSF is composed of a 50 kDa ligand binding α subunit (GM-CSFRα) and a 120 kDa signal transduction βC subunit. The functional GM-CSF receptor takes on a 2:2:2 complex of GM-CSF, GM-CSFRα, and βC. It should be noted that βC subunit is also part of the receptor complex for IL-3 and IL-5, which may signal through GM-CSFRα and βC. Incorporating Syndecan-2 as a co-receptor is another way GM-CSF can be utilized.

GM-CSF has many features.It induces the production of monocytes, neutrophils and eosinophils in CD34 + stem cell precursors.It cooperates with IL-4 or FLT-3 ligand to induce the development and maintenance of bone marrow and dermal dendritic cells.It also ACTS as a chemoattractant of neutrophil and dendritic cell. GM-CSF promotes Th1 and Th17 cell-mediated autoimmune inflammation and inflammatory activation of dendritic cells, microglia, alveolar macrophages, and eosinophils.In addition, it collaborates with G-CSF to promote tumor cell proliferation and invasion.
