Testing Principle

The quantitative sandwich enzyme immunoassay technique is utilized in this kit to detect rat IL-4 levels. Initially, a high affinity plate is pre-coated with a specific anti-rat IL-4 antibody. Next, test samples, standards, and biotinylated detection antibodies are added to the wells of the plate. These components combine with the solid-phase antibodies and detection antibodies to produce immune complexes after incubation. Unbound material is removed through a washing step prior to the addition of horseradish peroxidase-labeled streptavidin (HRP). A chromogenic substrate is then introduced, and the solution is incubated in the dark. The reaction is stopped by adding a stop solution, and the absorbance is measured at a wavelength of 450 nm with a reference correction wavelength of either 540 nm or 570 nm.

Background: IL-4

IL-4 is a type of secretory protein that consists of 129 amino acids. This cytokine is primarily produced by activated T lymphocytes, mast cells, and basophils. IL-4 is known for its ability to regulate various immune responses by acting on different types of cells. Its functions include stimulating the activation of B cells as well as inducing the expression of class II MHC molecules in quiescent B cells. DNA synthesis is also impacted by IL-4 as it is known to be a common irritant. This cytokine can increase the secretion and cell surface expression of IgE and IgG1. It also plays a vital role in regulating the expression of low-affinity Fc receptors for IgE (CD23) on various immune cells like lymphocytes and monocytes. Moreover, IL-4 positively regulates the expression of IL31RA in macrophages.
