Testing Principle：

In this assay kit, the double antibody sandwich ELISA method is utilized. The conjugate plate is pre-coated with a specific anti-human GDF-15 Capture Antibody that has high affinity. The process involves the sequential addition of the standard substance, the sample to be tested, and the detection antibody labeled with biotin into the HRP-plate well. After thorough oscillation and mixing at room temperature, the GDF-15 in the sample binds with both the solid phase antibody and the detection antibody during a 2-hour incubation period. Subsequently, the well is extensively washed to eliminate any free or unbound components. Next, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is introduced. Following another round of washing, TMB color substrate is added and incubated in darkness at room temperature to promote color development. The intensity of the color reaction is directly proportional to the concentration of GDF-15 present in the sample. To stop the reaction, a termination solution is added, and the absorbance value is measured using a microplate analyzer at a detection wavelength of 450 nm (with a correction wavelength range of 570-630 nm).

Background：

RAGE, also known as advanced glycation end product receptor, is a transmembrane immunoglobular receptor family that weighs around 35kDa. Its name is derived from its capability to bind to advanced glycation end products. RAGE is categorized as a recognition receptor due to its role in innate immunity and its ability to identify ligands through a shared motif. Its involvement has been identified in several chronic diseases including atherosclerosis, peripheral vascular disease, myocardial infarction, congestive heart failure, diabetes, and Alzheimer's disease. Interestingly, RAGE expression is highest in the lungs, particularly in type I alveolar epithelial cells. However, in idiopathic pulmonary fibrosis (IPF), RAGE expression is notably absent, indicating differential regulation and expression patterns between the lung and cardiovascular systems. Notably, when RAGE is blocked or knocked out, it hampers cell adhesion while promoting cell proliferation and migration.
