Testing Principle：

The double antibody sandwich ELISA kit operates by utilizing the principle of specific antibody binding to detect GDF-15 levels. The capture antibody, targeted towards human GDF-15, is pre-coated onto a conjugate plate with a high affinity surface. To perform the assay, standard substances, test samples, and biotin-labeled detection antibody are sequentially added to the wells containing the conjugate plate. Under room temperature conditions, the GDF-15 in the samples interacts with both the solid phase capture antibody and the detection antibody after thorough mixing and oscillation during a 2-hour incubation process.
After a thorough washing step to remove any unbound components, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is introduced. Following another round of washing, a TMB color substrate is added to the wells, and the plate is incubated in darkness at room temperature to allow color development. The intensity of the color reaction is directly proportional to the concentration of GDF-15 in the samples. To stop the reaction, a termination solution is added, and the absorbance value is measured at a detection wavelength of 450 nm using a microplate analyzer (with a correction wavelength set between 570-630 nm).

B cell activating factor (BAFF), also known as B lymphocyte stimulating factor (BLYS) or tumor necrosis factor ligand superfamily member 13B(TNFSF13B), has a vital role in the proliferation and differentiation of B cells. It serves as an effective activator, specifically in B cell line cells. BAFF acts as an immune stimulant, playing a crucial role in maintaining normal immunity. When BAFF is inadequately expressed, B cell activation is hindered, resulting in insufficient production of immunoglobulin and subsequent immunodeficiency. On the other hand, excessive production of BAFF can lead to an overabundance of antibodies, which can cause systemic lupus erythematosus, rheumatoid arthritis, and other autoimmune diseases. Overall, the expression of BAFF is crucial for the proper functioning of the immune system.
