Principle of Detection: The detection principle employed in this kit is double antibody sandwich enzyme-linked immunosorbent assay (ELISA) technology. A high-affinity ELISA plate is pre-coated with a specific anti-mouse CXCL10/IP-10/CRG-2 antibody. The wells of the enzyme-labeled plate are then filled with standards, test samples, and biotinylated detection antibody. Following incubation, the CXCL10/IP-10/CRG-2 in the sample binds to both the solid-phase antibody and the detection antibody, forming an immune complex. Unbound substances are removed through washing, and horseradish peroxidase-labeled streptavidin (Streptavidin-HRP) is added. After another round of washing, a chromogenic substrate is added to develop color. The reaction is stopped with a stop solution, and the absorbance value is measured at a wavelength of 450 nm, with reference correction wavelengths at 540 nm or 570 nm.

Background: CXCL10, also known as 10 kDa interferon-γ inducible protein (β-IP10 or IP-10), is a small cytokine belonging to the CXC chemokine family. It is secreted by various cell types, including monocytes, endothelial cells, and fibroblasts, in response to IFN-α. CXCL10 has multiple effects, such as attracting monocytes and T cells, promoting the adhesion of T cells and endothelial cells, exhibiting anti-tumor activity, and inhibiting bone marrow colony formation and angiogenesis. CRG-2, the mouse homolog of human IP-10, shares approximately 67% amino acid sequence identity with human IP-10.
