Testing Principle：In this ELISA kit, the microplates are coated with monoclonal antibodies against human and mouse COX-2, forming a double antibody sandwich setup. When the sample or standard containing COX-2 is added, it binds to the antibody fixed on the plate, while the unbound components are removed through washing. To further enhance the detection, biotin-labeled polyclonal antibodies against human and mouse COX-2 are introduced, which specifically bind to the COX-2 already attached to the microplates. After washing away the excess unbound components, a Streptavidin-labeled horseradish peroxidase is introduced to form a complex, exploiting the specific interaction between Streptavidin and biotin. By removing the unbound reagents through washing, a substrate solution is added, initiating a color change in the solution, gradually turning it from colorless to blue. The color change process is stopped by adding a stop solution, resulting in a yellow color. Finally, the absorbance of the yellow solution is measured using a microplate analyzer to quantify the amount of COX-2 present.
