Testing Principle

For this experiment, a double antibody sandwich ELISA was utilized. The microplate was coated with an anti-human OPG monoclonal antibody, which allowed for the binding of OPG present in both the sample and the standard. After the free components were removed by washing, a biotin-labeled anti-human OPG polyclonal antibody was added, which further combined with the OPG bound on the microplate. The free components were washed away once again. To form the complex, streptavidin-labeled horseradish peroxidase was added, which specifically recognized biotin. After washing away any unbound components, a substrate solution (chromogenic agent) was added, which caused the color of the solution to gradually change to blue. Finally, a stop solution was added to halt this changing process and turn the solution yellow. Using a microplate analyzer, the absorbance was measured.
