Description
JC-10, a commonly used fluorescent probe, offers a reliable method to detect changes in mitochondrial membrane potential △Ψm. It is applicable for detecting mitochondrial membrane potential in cells, tissues, and purified mitochondria. The principle behind JC-10's detection mechanism lies in its ability to aggregate and form polymer structures within the mitochondrial matrix when the membrane potential is high. Under these conditions, JC-10 exhibits red fluorescence. Conversely, when the membrane potential is low, JC-10 remains in a monomeric state, resulting in green fluorescence.
The convenient fluorescence color change allows for the detection of alterations in mitochondrial membrane potential. The relative ratio of red and green fluorescence is often used to quantify mitochondrial depolarization. Notably, the decline in mitochondrial membrane potential serves as a crucial indicator of the early stages of apoptosis, or programmed cell death. By monitoring the transition of JC-10 from red fluorescence to green fluorescence, it becomes feasible to easily detect the reduction in cell membrane potential. Thus, the conversion of JC-10's red fluorescence to green fluorescence serves as an early detection marker for apoptosis.
JC-10 monomers have a maximum excitation wavelength of 515nm and emit light at 529nm, while JC-10 polymers have a maximum excitation wavelength of 585nm and emit light at 590nm. To observe the red and green fluorescence efficiently, conventional settings for these wavelengths are recommended for practical use.
