TUNEL Apoptosis Detection Kit-DAB

An identifying characteristic of apoptosis is the degradation of chromosomal DNA in cells, which commonly occurs through a specific and regular process. This produces DNA fragments of various lengths that typically correspond to integer multiples of 180 bp-200 bp, resulting in a distinctive ladder-like pattern when observed through agarose gel electrophoresis. The TUNEL method is employed by this kit, which relies on terminal deoxyribonucleotide transferase (TdT) to catalyze the incorporation of biotin-labeled dUTP onto the 3´-OH ends of fragmented DNA in apoptotic cells. The resulting biotin-DNA is then detected by binding to horseradish peroxidase (HRP)-labeled streptavidin, allowing for visual detection of apoptotic cells via DAB catalyzed by HRP. Unlike normal or proliferating cells, which tend to have few DNA breaks and therefore do not react to the TUNEL method, apoptotic cells can be selectively targeted and counted using this technique. Additionally, because this method can directly identify cells undergoing apoptosis in situ, it offers a faster and more efficient detection method compared to alternatives that may identify necrotic cells or those with DNA strand breaks induced by radiation or drugs. The kit includes several key components, such as Biotin TUNEL Reaction Buffer, BTdT enzyme, Streptavidin-HRP, and HProteinase Ku, among others.

The recommended storage conditions for this product are as follows: it should be kept at -20° C and away from light. Similarly, the TUNEL Reaction Buffer should also be stored at -20° C and away from light to prevent damage caused by repeated freezing and thawing. It is important to adhere to these storage requirements to maintain the quality of the product. When stored under the recommended conditions, this product can maintain its effectiveness for a period of 12 months.
