Mitochondrial Membrane Potential Assay Kit(JC-1)

The JC-1 mitochondrial membrane potential detection kit is an excellent tool for monitoring changes in cell, tissue, or purified mitochondrial membrane potentials. It utilizes JC-1 as a fluorescent probe, enabling rapid and sensitive detection of ΔΨm alterations. JC-1 is a popular fluorescent probe that can detect mitochondrial membrane potentials with great accuracy. When the mitochondrial membrane potential is high, JC-1 polymerizes and aggregates in the matrix of the mitochondria, generating red fluorescence. In contrast, when the mitochondrial membrane potential is low, JC-1 cannot accumulate in the matrix, resulting in green fluorescence. This is an extremely convenient approach for detecting changes in mitochondrial membrane potential by analyzing fluorescence color. The ratio of red and green fluorescence is used to determine the extent of mitochondrial depolarization.
Early-stage apoptosis is often accompanied by a decrease in mitochondrial membrane potential. JC-1 monomer transitioning from red fluorescence to green fluorescence can easily monitor the decline in cell membrane potential, making it an effective method for identifying apoptotic cells. The JC-1 monomer's maximum excitation wavelength is 515nm, and its maximum emission wavelength is 529nm. In contrast, the maximum excitation wavelength of JC-1 polymer is 585nm, with a maximum emission wavelength of 590nm. It is recommended to use conventional settings to observe red and green fluorescence.
The kit includes CCCP as a positive control to stimulate a decrease in mitochondrial membrane potential. A six-well plate's samples will generate a total of 100 samples, while a 12-well plate will generate a total of 200 samples. The kit comprises JC-1 (200×) in five tubes of 100μl each, ultrapure water (90mL), JC-1 staining buffer (5×) (80mL), and CCCP (10mM) (20μl).

Ensure that the product is stored in a dark environment at a temperature of -20°C. It is advisable to refrain from freezing and thawing the product repeatedly for a duration of one year.
