In a normal cell cycle, the different phases have distinct DNA content: G0 and G1 phases have one set of chromosomes, G2 and M phases have two sets of chromosomes, and the S phase is the phase where DNA replication occurs. By staining cells with PI, the fluorescence intensity can vary, allowing differentiation between cells in different phases of the cell cycle. For instance, cells in G0 and G1 phases exhibit a fluorescence intensity of 1, while cells in G2 and M phases have a fluorescence intensity of 2. Cells in the S phase will display a fluorescence intensity somewhere between 1 and 2. Moreover, PI staining can also be utilized to detect apoptosis, a process characterized by cell death. During apoptosis, the nucleus shrinks and DNA fragments, resulting in the loss of DNA from the permeable cell membrane during staining. Consequently, when analyzed by flow cytometry, the fluorescence intensity of apoptotic cells will be less than 1, forming a distinct sub-G1 peak. Another way to measure apoptosis is by examining changes in light scattering properties of cells through flow cytometry. In the early stages of apoptosis, chromatin condenses, leading to an increase in cell density and a noticeable decrease in forward angular light scattering. Later in the apoptotic process, the formation of apoptotic bodies occurs, bringing about a significant reduction in both forward and lateral angular light scattering. The components included in this product are Propidium Iodide (500 μl), BRNase A (500 μl), CStaining Buffer (25ml), in a total volume of 50TA. By using this kit, researchers can accurately analyze cell cycle progression and apoptosis based on the fluorescence and scattering properties of stained cells. |
