Caspase-1, also known as interleukin 1β converting enzyme (ICE), is an enzyme involved in the processing and activation of pro-inflammatory cytokines. This enzyme plays a critical role in the innate immune response and is responsible for the release of mature IL-1β and IL-18 from their inactive precursors. The Caspase 1 Activity Detection Kit enables the detection of caspase 1 enzyme activity in cell or tissue lysate through spectrophotometry. A member of the caspase family, caspase-1 plays a crucial role in cell apoptosis, and is the only caspase capable of cleaving IL-1b and IL-18 precursors to produce active cytokines. This kit utilizes caspase-1 to catalyze the substrate Ac-YVAD-pNA to produce yellow pNA, which can be measured through absorbance, allowing for the detection of caspase 1 activity. The pNA has a strong absorption near 405nm, which makes it easily detectable. The kit includes lysis buffer, assay buffer, Ac-YVAD-pNA (2mM), and pNA (10mM), which allows for the testing of up to 20 or 100 samples in addition to the standard curve using a microplate reader or a spectrophotometric test cup. This detection kit is useful for studying the regulation of cell apoptosis through the cleavage of Bcl-XL and the immune inflammatory response mediated by related cytokines.

The recommended storage temperature for Ac-YVAD-pNA and pNA is -20°C. It is important to store them in a dark environment to maintain their integrity. These compounds have a validity period of 12 months, meaning they can be used effectively within this time frame.
