Testing Principle
The Mouse VEGF ELISA Kit employs a quantitative sandwich enzyme immunoassay method that utilizes a plate coated with a specific anti-mouse VEGF antibody of high affinity. Test samples, standards, and biotinylated detection antibodies are added to the wells of the enzyme-labeled plate, and after incubation, the solid-phase antibodies and detection antibodies combine with the VEGF present in the samples to form immune complexes. Unbound material is removed through washing, and horseradish peroxidase-labeled streptavidin (HRP) is introduced. Following another washing step, the addition of a chromogenic substrate results in color development in the dark. The stop solution is added to halt the reaction, and absorbance is measured at a wavelength of 450 nm (with reference correction wavelength of 540 nm or 570 nm).

Vascular permeability factor or vascular endothelial growth factor (VEGF or VEGF-A) serves as a crucial regulator of angiogenesis in both fetuses and adults. Within the PDGF family, VEGF exhibits a unique characteristic of eight conserved cystine residues in its cystine junctions structure, forming dimers held together by reverse parallel disulfide bonds. This remarkable arrangement distinguishes VEGF from other proteins in the same family and underscores its significance in stimulating blood vessel growth.
The expression of human VEGF can be manipulated to produce different subtypes through selective cleavage and varying amino acid sequence lengths. These subtypes include VEGF121, VEGF145, VEGF165, VEGF183, VEGF189, and VEGF206. Among these subtypes, VEGF165 is the most abundant, while VEGF121 and VEGF189 follow closely behind. However, all subtypes, except VEGF121, share a basic heparin-binding region and are unable to diffuse freely. Hence, the expression and manipulation of VEGF subtypes could lead to the development of targeted therapies for various diseases.
VEGF165, a crucial protein involved in various cellular processes, exhibits a remarkable 88% homology in its amino acid sequence between humans and mice and rats. Its wide-ranging expression is observed in numerous cells and tissues, such as skeletal muscle cells, cardiomyocytes, hepatocytes, osteoblasts, neutrophils, macrophages, keratinocytes, brown fat cells, CD34+ stem cells, endothelial cells, fibroblasts, and vascular smooth muscle cells. The regulation of VEGF expression is complex and multifaceted. It can be induced by hypoxia as well as cytokines like IL-1, IL-6, IL-8, tumor suppressor M, and tumor necrosis factor α. Importantly, VEGF expression also varies during different stages of development and into adulthood. These findings underscore the significance of VEGF in various biological processes and highlight its intricate modulation under diverse physiological conditions.
The dimer of VEGF binds to two related tyrosine kinase receptors, which are VEGF R1 (also known as FLT-1) and VEGF R2 (FLK-1 /KDR).VEGF can also induce the homodimer and autophosphorylation of the latter. These receptors have seven extracellular immunoglobulin domains and one intracellular separate tyrosine receptor domain.Vascular endothelial cells and some other non-endothelial cells express VEGF receptors. Although VEGF and R1 have the highest affinity, VEGF R2 is the major factor in the regulation of VEGF angiogenesis. VEGF165 also binds to the brachioplatin (Semaporin) receptor, Neuropilin-1, thereby promoting complex formation with VEGF R2.
