Testing Principle

The technique employed in this kit is the quantitative sandwich enzyme immunoassay, utilizing a high affinity plate coated with a specific anti-mouse IL-10 antibody. To initiate the assay, standards, test samples, and biotinylated detection antibodies are introduced into the wells of an enzyme-labeled plate. Following a period of incubation, the IL-10 in the samples binds to both the solid-phase antibodies and the detection antibodies, generating immune complexes. Subsequent washing removes any unbound material, after which horseradish peroxidase-labeled streptavidin (HRP) is added. Another round of washing ensues, and then a chromogenic substrate is introduced to induce color development in the absence of light. The reaction is halted by adding a stop solution, and the resulting absorbance is measured at a wavelength of 450 nm, with the reference correction wavelength being 540 nm or 570 nm.

Background: IL-10

Interleukin-10 (IL-10) is a member of the IL-10α helix family, which includes IL-19, IL-20, IL-22, IL-24, and IL-26/AK155. It is also known as the cell growth factor synthesis inhibitor (CSIF). IL-10 is produced by various activated hematopoietic cells, hepatic stellate cells, keratinocytes, and placental trophoblast cells. Interestingly, human IL-10 is biologically active in mouse cells, whereas mouse IL-10 does not exhibit activity in human cells. When comparing amino acid sequences, mature human IL-10 shares 86% homology with horse IL-10 and 72-80% homology with bovine, dog, cat, guinea pig, mouse, sheep, pig, and rat IL-10. This cytokine forms non-covalently bound homologous dimers, characterized by two in-chain disulfide bonds. Its molecular weight is approximately 36kDa.

The biological activity of IL-10 is achieved through the allodimer complex composed of the IL-10 receptor subunit α and β of type II cytokine. IL-10 receptor subunit α is a transmembrane glycoprotein with a molecular weight of 110kDa that is mainly expressed in lymphocytes, NK cells, macrophages, monocytes, astrocytes, intestinal epithelial cells, trophoblastic cells, and activated hepatic stellate cells. On the other hand, receptor β, with a molecular weight of 75kDa, is widely expressed.
The IL-10 dimer binds two IL-10 Rα chains, causing the addition of two IL-10 Rβ chains. While IL-10 Rβ is required for signal transduction, it does not bind IL-10 directly. Furthermore, IL-10 Rβ forms receptor complexes with IL-20 Rα, IL-22 Rα1, and IL-28 Rα, leading to the formation of IL-22, IL-26, IL-28, and IL-29.

IL-10 is involved in immune regulation including inhibition and stimulation. It promotes the anti-inflammatory effect of M2-type macrophages by inhibiting the amplification and activation of Th1 and Th17 cells. The regulation of IL-10 expression by immunomodulatory T cells (Treg) and regulatory B cells is important for Treg proliferation. However, IL-10 inhibited the proliferation of Treg and myeloid cells in the tumor environment.IL-10 induces accumulation and activation of CD8 T cells in tumors.IL-10 plays a protective role by limiting tissue damage in arthritis and promoting regeneration after muscle damage, but it may contribute to the persistence of viral infections.IL-10 levels were elevated in Sjogren's syndrome (salivary), primary central nervous system lymphoma (cerebrospinal fluid), and ovarian cancer (serum and ascites).While the level in serum of patients with recurrent heart disease or pre-xian and semen of male infertility were decreased.
