Mouse IFN gamma ELISA Kit


Testing Principle

This enzyme immunoassay kit employs a quantitative sandwich technique to measure mouse IFN-gamma levels. The high affinity plate comes pre-coated with a specific anti-mouse IFN-gamma antibody. Test samples, detection antibodies, and standards are pipetted into the enzyme-labeled wells of the plate. Following incubation, the present IFN-gamma combines with the solid-phase antibodies along with the detection antibodies.
Any unbound material is removed by washing, and then horseradish peroxidase-labeled streptavidin is added. The reaction is then incubated again before washing and the addition of a chromogenic substrate. The color is then developed in the dark before a stop solution is added, and absorbance is measured using a reference correction wavelength of either 540 nm or 570 nm at a wavelength of 450 nm.

Background: IFN-gamma

Type II interferon, also known as interferon gamma, is a crucial cytokine that can modulate the immune system. Its antiviral properties led to its discovery and it is now recognized for its pivotal role in host defense. In addition to its antiviral effects, this cytokine also displays antiproliferative and immunomodulatory functions.

Interferon gamma is known for its ability to induce the production of cytokines and to upregulate the expression of various membrane proteins in a range of cell types. These include but are not limited to MHC-I, MHC-II, Fc receptors, leukocyte adhesion molecules, and B7 family antigens. Moreover, interferon gamma has been found to activate macrophages and guide the synthesis, type transformation, and secretion of B-cell immunoglobulin. It also has a significant impact on T helper cell subtypes, inhibiting the differentiation of Th2 cells and promoting the growth of Th1 cells. Notably, interferon gamma is involved in several inflammatory diseases, such as autoimmunity and atherosclerosis.

Interferon gamma is a dimer with a molecular weight of 20-25 kD, and is glycosylated to varying degrees. In humans, the amino acid sequence of Interferon gamma is highly homologous to that of rhesus monkeys, with a close to 90% similarity. The homology with Interferon gamma in cattle, dogs, horses, cats and pigs ranges from 59-64%. Meanwhile, Interferon gamma in cotton mice and rats has a lower homology of only around 37-43%. This demonstrates the remarkable variability of Interferon gamma across different species, despite its overarching biological function.

The interferon dimer first binds to the transmembrane receptor IFN-γ RI (α subunit) and then to IFN-γ RII (β subunit) to form a functional complex consisting of two α subunits and two β subunits.IFN-γ RII in the complex receptor can increase ligand affinity and signal transduction efficiency.Although gamma subunits are constitutively expressed in many cell types, the regulatory state of gamma interferon, the receptor expressed by beta subunits, is tightly regulated.

Interferon gamma is secreted by many cells under a range of inflammatory conditions, including dendritic epidermal/γδ T cells, keratinocytes, peripheral bloodγδ T cells, mast cells, neurons, CD8+T cells, macrophages, B cells, neutrophils, natural killer cells, CD4+T cells, and testicular sperm cells.
