Testing Principle

The sandwich enzyme immunoassay technique is employed in this kit, utilizing a quantitative approach. Initially, a plate with high affinity is coated with a specific anti-mouse CCL2 antibody. Subsequently, the wells of the plate are filled with standards, test samples, and biotinylated detection antibodies. Following an incubation period, the CCL2 present in the samples interacts with both the solid-phase antibodies and the detection antibodies, resulting in the formation of immune complexes. Subsequent washing eliminates any unbound material, after which horseradish peroxidase-labeled streptavidin (HRP) is added. Further washing is performed, and a chromogenic substrate is introduced, leading to the development of color in a dark environment. The reaction is halted by introducing a stop solution, and the absorbance is measured at a wavelength of 450 nm (taking into account a reference correction wavelength of either 540 nm or 570 nm).
