Testing Principle

This enzyme immunoassay kit employs a quantitative sandwich technique, where a highly specific anti-mouse GM-CSF antibody is already coated on a plate with high binding affinity. The test samples, along with standards and biotinylated detection antibodies, are added to the wells of the enzyme-linked plate. The GM-CSF present in the samples interacts with the solid-phase antibodies, as well as those in the detection antibodies, forming immune complexes. Unbound material is then removed by washing, and horseradish peroxidase-labeled streptavidin (HRP) is added. In the next step, a chromogenic substrate is added to the mixture, and color development occurs in the dark. To stop the reaction, a stop solution is added, and absorbance is measured at a nominated wavelength of 450 nm (with reference correction wavelength of either 540 nm or 570 nm). Overall, the process is designed to allow for the quantitative measurement of GM-CSF in a given sample.

Background:GM-CSF

GM-CSF, or CSF-2 for short, is a versatile cytokine belonging to the common β chain (βC) family of proteins. With a molecular weight of 30 kDa, GM-CSF adopts a α-helix conformation featuring a pair of in-chain disulfide bonds. This protein is primarily produced by immune cells, as well as various types of mesenchymal and epithelial cells. Secreted in the form of various glycosylated monomers, GM-CSF plays a crucial role in promoting the growth and maturation of a range of blood cells. Both IL-3 and IL-5 are also members of the βC family of cytokines.

GM-CSF, also known as granulocyte-macrophage colony-stimulating factor, plays a crucial role in inflammation by upregulating various cell types. These include encephalitis causing T cells, allergen-exposed lung endothelial cells, and IgE activated mast cells. In terms of structural similarity, the mature human GM-CSF shares 54% and 63% amino acid sequence identity with mouse and rat GM-CSF, respectively. The receptor for GM-CSF is composed of a 50 kDa ligand binding α subunit (GM-CSFRα) and a 120 kDa signal transduction βC subunit. Studies have reported that the functional GM-CSF receptor exists as a 2:2:2 complex of GM-CSF, GM-CSFRα, and βC. Additionally, the βC subunit is shared by the receptor complex of IL-3 and IL-5, indicating potential cross-signaling. Furthermore, GM-CSF can utilize Syndecan-2 as a co-receptor, expanding its signaling capabilities.

GM-CSF, also known as granulocyte-macrophage colony-stimulating factor, possesses various important functions. One of its primary abilities is to stimulate the production of monocytes, neutrophils, and eosinophils from CD34+ stem cell precursors. Additionally, GM-CSF acts together with IL-4 or FLT-3 ligand to support the development and maintenance of bone marrow and dermal dendritic cells. Moreover, it serves as a chemoattractant for neutrophils and dendritic cells, guiding them towards specific locations. Notably, GM-CSF plays a significant role in promoting Th1 and Th17 cell-mediated autoimmune inflammation, as well as inflammatory activation of dendritic cells, microglia, alveolar macrophages, and eosinophils. Furthermore, it collaborates with G-CSF, another type of colony-stimulating factor, to enhance tumor cell proliferation and invasion. These various features highlight the critical contributions of GM-CSF to immune responses, inflammation, and cellular development.
