Human IL-17/IL-17A ELISA Kit

Testing Principle

This assay kit utilizes a quantitative sandwich enzyme immunoassay technique to detect human IL-17/IL-17A. The solid-phase plate is pre-coated with a specific anti-human IL-17/IL-17A antibody with high affinity. To initiate the assay, the samples, standards, and biotinylated detection antibodies are added to the wells of the labeled plate. After an incubation period, immuno complexes are formed by the combination of the present IL-17/IL-17A with the solid-phase and detection antibodies. Excess unbound materials are removed through a washing process, followed by the addition of horseradish peroxidase-labeled streptavidin (HRP). Subsequently, a chromogenic substrate is introduced to develop a color in the dark. The reaction is stopped by adding a stop solution that allows for the measurement of absorbance at 450 nm with a reference correction wavelength of 540 nm or 570 nm.

Background: IL-17

The IL-17 family consists of six cytokines that share a similar cysteine junction structure but differ at the N-terminal. These glycoproteins are secreted as dimers and induce local cytokine production, attracting granulocytes to inflammatory sites. IL-17 is primarily induced by IL-17 and IL-23 in activated CD4+ T cells, in contrast to Th1 or Th2 cells. Furthermore, only IL-23 can induce IL-17F, which is most similar to IL-17 but only in activated monocytes. On the other hand, IL-17B binds to the IL-17B receptor, not the IL-17 receptor, and is most related to IL-17D, expressed in resting CD4+ T cells and CD19+ B cells. Th2 cells mainly produce IL-17E, reminiscent of eosinophil recruitment to lung tissue. IL-17C expression is very limited, with detection in adult prostate and fetal kidney Banks.
