Testing Principle
The Mouse IL-6 ELISA Kit employs a quantitative sandwich enzyme immunoassay method, which includes pre-coating a high affinity plate with a specific anti-mouse IL-6 antibody. Next, biotinylated detection antibodies, standards, and test samples are sequentially added to the wells of the enzyme-labeled plate. Following an incubation period, immune complexes are formed by the combination of IL-6, the solid-phase antibodies, and the detection antibodies. Any unbound material is removed through washing, and then horseradish peroxidase-labeled streptavidin (HRP) is added. After additional washing, a chromogenic substrate is introduced. The color is developed in a dark environment and terminated by adding a stop solution. Finally, the absorbance is determined at 450 nm while using a reference correction wavelength of either 540 nm or 570 nm.

Mature human IL-6 is made up of 183 amino acids and has 39% sequence identity with mouse and rat IL-6.
