Testing Principle
This quantitative sandwich enzyme immunoassay kit utilizes a specific anti-mouse IL-2 antibody that is pre-coated on a high affinity plate. The enzyme-labeled plate contains wells in which standards, test samples, and biotinylated detection antibodies are added. Following an incubation period, the IL-2 present in the samples combines with the solid-phase antibodies and the detection antibodies, forming immune complexes. Unbound material is then washed away, and horseradish peroxidase-labeled streptavidin (HRP) is added. After another round of washing, a chromogenic substrate is introduced, resulting in the development of color in a dark environment. To stop the reaction, a stop solution is added, and the absorbance is measured at a wavelength of 450 nm (with a reference correction wavelength of either 540 nm or 570 nm).

Interleukin-2 (IL-2), also referred to as T cell growth factor (TCGF), can be categorized as a member of the common γ chain (γc) cytokine family. This glycosylated α-helical polypeptide possesses a molecular weight within the range of 15-18 kDa. As a monomeric protein, IL-2 exhibits a rather short half-life of less than 30 minutes. The human IL-2 precursor encompasses a total of 153 amino acids, inclusive of a 20-amino acid signal sequence and a 133-amino acid mature polypeptide. Remarkably, the mature form of IL-2 features a threonine O-glycosylated site, along with three cysteine residues. Notably, two of these cysteine residues are capable of establishing an indispensable in-chain disulfide bond that contributes to the activity of IL-2. To summarize, IL-2, also known as T cell growth factor, is a glycosylated α-helical polypeptide belonging to the γc cytokine family. Its monomeric structure with a short half-life, along with the presence of a threonine O-glycosylated site and cysteine residues, contributes to its biological activity.
The level of homology between human mature IL-2 and other species varies, with the highest homology being observed in macaques at 97%. Nonetheless, despite only 60% homology in mice, human IL-2 is still biologically active in mouse cells. Multiple cell types have been reported to secrete IL-2, including γδ T cells, activated CD4+ and CD8+ T cells, microglia, hematopoietic stem cells, and even neurons.
The IL-2 receptor, known as IL-2R, is composed of three subunits: a 55 kDa CD25/IL-2R α chain, a 70 kDa CD122/IL-2R β chain, and a 65 kDa CD132/γ chain. The binding of IL-2 to CD25 forms a disubunit complex, which then recruits CD122 and CD132 to create a signal complex consisting of four subunits. Interestingly, CD122/IL-2Rβ can also form a four-subunit signaling complex with IL-15, in addition to its interaction with IL-2. Furthermore, CD132/γc acts as a receptor for various cytokines, including IL-4, IL-7, IL-9, IL-15, and IL-21. These cytokines can bind to CD132/γc and initiate signaling pathways. In vivo, IL-2 is the key to the development, maintenance and function of regulatory T cells (Treg), which provide protection against autoimmune diseases. In addition, IL-2 also promotes autoimmune inflammation of target organs by regulating the expression of trafficked genes in T cells and the production of Th2 cytokines. In CD8+ T cell subsets, IL-2 is essential for optimal primary response and differentiation to terminal effector cells.IL-2 also promotes the development and activation of CD8+ T cells into memory cells.
