Testing Principle

The sandwich enzyme immunoassay technique is employed in this kit, utilizing quantitative measurement. The plate is coated with a specific anti-mouse IL-12 p70 antibody that exhibits high affinity. To initiate the assay, standards, test samples, and biotinylated detection antibodies are introduced into the wells of the enzyme-labeled plate. Following an incubation period, the IL-12 p70 present in the samples binds with the solid-phase antibodies and the detection antibodies, resulting in the formation of immune complexes. Unbound material is eliminated through a washing step, after which horseradish peroxidase-labeled streptavidin (HRP) is added. Another wash is performed, followed by the addition of a chromogenic substrate, which develops color in the absence of light. To halt the reaction, a stop solution is introduced, and the absorbance is measured at a wavelength of 450 nm (with a reference correction wavelength of either 540 nm or 570 nm).

Interleukin-12 p70 (IL-12) is a versatile cytokine known by various names such as natural killer cell stimulating factor (NKSF) or cytotoxic lymphocyte maturation factor (CLMF). It is a heterodimeric molecule comprising 40kDa (p40) and 35kDa (p35) subunits. These subunits can interact in different ways to form different complexes. Human p40 can exist as a monomer or homodimer, or it can bind with p35 to form IL-12. Additionally, it can bind with p19 to form the IL-23 complex. Interestingly, both the homologous dimer p40 and IL-23 can bind to the IL-12 receptor as antagonists without triggering any signaling. It is worth noting that the IL-12 p40 subunit is also shared with another cytokine called IL-23, which exhibits similar yet distinct biological activities compared to IL-12. Macrophages and dendritic cells, monocytes, Langerhans cells, neutrophils, keratinocytes, plasmacytoid dendritic cells, microglia, CD8+DC (in mice), and non-germinal centers (CD38-CD44+) B cells (in humans) are known to produce IL-12. This cytokine plays a crucial role in enhancing cytotoxic activity and stimulating the production of interferon γ (IFN-γ) in various immune cells such as NK cells, T cells, and dendritic epidermal T cells. Furthermore, IL-12 has been observed to induce the production of IFN-γ in macrophages as well.

The immune response driven by CD4+Th1 cells is promoted by a group of cytokines known as the IL-12 family, comprising IL-12, IL-23, and IL-27. In the presence of an infection, IL-27 is released initially, facilitating the conversion of Th0 to Th0/1 cells. Subsequently, IL-12 is secreted, leading to the production of Th1 effector cells. The collaboration between IL-12 and IL-18 results in the generation of Th1 memory cells derived from the effector cells, which are then activated by IL-23. It is important to note that while mouse IL-12 exhibits functionality in both mouse and human cells, human IL-12 is incapable of inducing activity in mouse cells.
