Mouse IL-17 ELISA Kit

Testing Principle

In this kit, the sandwich enzyme immunoassay technique is employed for quantitative analysis. Initially, a plate with a high affinity is coated with a specific anti-mouse IL-17/IL-17A antibody. Then, the wells of the plate are filled with standards, test samples, and biotinylated detection antibodies. The IL-17/IL-17A present in the samples binds with the solid-phase antibodies and the detection antibodies, forming immune complexes. Unbound materials are removed through a washing process. Next, horseradish peroxidase-labeled streptavidin (HRP) is introduced. After washing again, a chromogenic substrate is added, and the color develops in darkness. To halt the reaction, a stop solution is added, and the absorbance is measured at a wavelength of 450 nm (reference correction wavelength of 540 nm or 570 nm).

Background: IL-17

The IL-17 family encompasses several pro-inflammatory cytokines sharing a conserved cysteine junction structure but differing at the N-terminal. These glycoproteins are secreted as dimers and exert their effects by stimulating local cytokine production and recruiting granulocytes to sites of inflammation. IL-17 is primarily induced by IL-17 and IL-23 in activated CD4+ T cells, setting it apart from Th1 or Th2 cells. On the other hand, IL-17f shows the closest resemblance to IL-17 but is only triggered by IL-23 in activated monocytes. IL-17b binds to the IL-17B receptor and lacks affinity for the IL-17 receptor. It shares the highest homology with IL-17D, which is expressed by resting CD4+ T cells and CD19+ B cells. Th2 cells predominantly produce IL-17e, which recruits eosinophils to lung tissue. Lastly, IL-17c exhibits a restricted expression pattern, with known detection in the adult prostate and fetal kidney Banks. By rearranging and summarizing the given information, we can understand the various members of the IL-17 family and their roles in inflammation and immune response.
