Testing Principle

The quantitative sandwich enzyme immunoassay technique is utilized in this kit, which involves a pre-coated high affinity plate with specific anti-human IL-4 antibody. To test samples, standards, and biotinylated detection antibodies are added to the enzyme-labeled plate wells. Incubation allows the formation of immune complexes where the IL-4 present in the samples combines with the solid-phase and detection antibodies. Afterward, unbound material is removed through washing and horseradish peroxidase-labeled streptavidin (HRP) is added. The addition of a chromogenic substrate then produces color in the dark. To stop the reaction, a stop solution is added, and the absorbance is measured at a wavelength of 450 nm with a reference correction wavelength of 540 nm or 570 nm.
