Testing Principle

The method utilized in this kit is a quantitative sandwich enzyme immunoassay employing a high affinity plate pre-coated with a specific anti-human IL-12 p70 antibody. To perform the assay, standards, test samples, and biotinylated detection antibodies are added to the wells of an enzyme-labeled plate. Following an incubation period, the IL-12 p70 present in the samples binds to the solid-phase antibodies and the detection antibodies, forming immune complexes. Unbound material is subsequently washed away, and a horseradish peroxidase-labeled streptavidin (HRP) is introduced. After another washing step, a chromogenic substrate is added to initiate color development in the absence of light. To terminate the reaction, a stop solution is added, and the resulting absorbance is measured at a wavelength of 450 nm (with reference correction at either 540 nm or 570 nm).

IL-12, also known as NKSF or CLMF, is a pluripotent cytokine composed of p40 and p35 subunits. The p40 subunit can exist as a monomer, homodimer, or combine with p35 or p19 to form IL-12 or IL-23. Both p40 homodimer and IL-23 can act as non-signaling antagonists by binding to IL-12R. Additionally, p40 can activate microglia and macrophages by binding to Rβ1. It is important to note that IL-12 shares the p40 subunit with IL-23, which exhibits similar yet distinct biological activities compared to IL-12.

IL-12, an immune regulatory cytokine, is produced by various immune cells including macrophages, dendritic cells, monocytes, Langerhans cells, neutrophils, keratinocytes, plasmacytoid dendritic cells, microglia, CD8+DC (in mice), and non-germinal centers (CD38-CD44+) B cells (in humans). One important function of IL-12 is its ability to enhance the cytotoxic activity of natural killer (NK) cells, T cells, and dendritic epidermal T cells. Additionally, IL-12 induces the production of interferon γ (IFN-γ) in these immune cells. It is worth mentioning that IL-12 has also been observed to stimulate IFN-γ production by macrophages.

IL-12, IL-23, and IL-27, which are members of the IL-12 family, play a crucial role in the development of the CD4+Th1 immune response. It is important to note that while mouse IL-12 exhibits activity in both human and mouse cells, human IL-12 does not demonstrate activity in mouse cells.
