Testing Principle：This ELISA kit makes use of a dual antibody sandwich technique. A specific anti-human IL-5 capture antibody is coated on a conjugate plate of high affinity. The test sample, the standard substance, and detection antibody labeled with biotin are added successively into the HRP-plate well. This mixture is then incubated at room temperature for a period of 2 hours while shaking constantly. During this incubation process, the IL-5 in the sample combines with the solid phase antibody and the detection antibody. After washing thoroughly to eliminate any free and unbound elements, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is introduced. After another washing cycle, a TMB color substrate is added and allowed to develop in the dark at room temperature. The intensity of the color reaction is associated with the concentration of IL-5 in the sample. To conclude the reaction, a termination solution is added, and the absorbance value is measured at a detection wavelength of 450 nm (correction wavelength 570-630 nm) using a microplate analyzer.

Background：

IL-5 is a cytokine belonging to the hematopoietic family, consisting of 115 amino acids. Unlike other cytokines, IL-5 is an activated glycoprotein and is a homodimer. It is produced by Th2 cells and mast cells and plays a critical role in Th2 cell activity. By binding to its receptor, IL-5 aids in the growth of B cells and promotes the secretion of immunoglobulin. Furthermore, it is a significant factor in the activation of eosinophils. Elevated levels of IL-5 have been linked to various ailments such as allergic rhinitis and asthma, while it has been found to be markedly decreased in sputum eosinophils. Also, IL-5 has been observed to be substantially elevated in circulatory and tracheal tissues.
