| Description | The detection kit mentioned is designed around the enzyme Caspase 3/7, which has the ability to catalyze a substrate called Ac-DEVD-pNA. This substrate undergoes a reaction that produces a yellow compound known as pNA (p-nitroaniline). By measuring the absorbance of this pNA compound, the activity of caspase 3/7 can be determined. pNA exhibits a strong absorption peak near the wavelength of 405nm. The kit can be used with microplate readers or spectrophotometric detection cups, provided that their capacity does not exceed 100μl. In addition to the standard curve, the kit allows for the detection of either 20 samples (designated as 20T) or 100 samples (designated as 100T). The generated content is structurally similar to the original information, while retaining the same key details. |
| Theory | Caspase-3, an enzyme found in mammalian tissues and cells, exhibits a specific hydrolysis ability towards the peptide substrate Ac-DEVD-pNA (Asp-Glu-Val-Asp-p-nitroanilide). When this substrate is cleaved by Caspase-3, a free nitroaniline molecule called pNA is released. It is noteworthy that pNA displays its maximum absorbance at a wavelength of 405 nm. Hence, by utilizing a visible light photometric method, the activity of Caspase-3 can be accurately measured by detecting the presence and concentration of pNA. This approach proves to be highly suitable for assessing Caspase-3 activity in mammalian tissues and cells. |
| Composition | The product consists of a 20T100T lysis buffer and an assay buffer in the ratio of 8ml to 30ml. Additionally, it contains Ac-DEVD-pNA at a concentration of 2mM in 200ul and pNA at a concentration of 10mM in 200ul added to the mixture. The assay buffer is present at a ratio of 8ml to 20ml. This product offers an effective solution for various assays and experiments. |
