Testing Principle：

This particular kit preferred to utilize a double antibody sandwich ELISA method, which involves incorporating a specific anti-human CXCL13 Capture Antibody that comes prepackaged on a high affinity conjugate plate. Firstly, a standard substance, as well as the sample to be tested, will be added to the HRP-plate well, followed by the biotin labeled detection antibody. After thorough oscillation and mixing, the sample will be allowed to undergo a 2-hour incubation process at room temperature, where the CXCL13 in the sample will be combined with the solid phase antibody and detection antibody. Once the free and unbound ingredients have been washed out completely, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) will be added. Another wash follows before TMB color substrate can be introduced and incubated in the dark at room temperature to facilitate color development. In measuring the CXCL13 concentration, the depth of color reaction will be positively correlated. The termination solution will work to end the reaction, and a microplate analyzer will measure the absorbance value at 450 nm detection wavelength (correction wavelength 570-630 nm).

Background：
