Testing Principle:
The double antibody sandwich ELISA method is employed in this kit. The conjugate plate is pre-coated with a specific anti-human CXCL13 Capture Antibody of high affinity. The procedure starts by adding the standard substance, the sample to be tested, and the biotin-labeled detection antibody into the HRP-plate well successively. Following thorough oscillation and mixing, the sample is incubated at room temperature for 2 hours to allow CXCL13 to bind to the solid phase antibody and the detection antibody. After thorough washing to remove any unbound components, horseradish peroxidase labeled streptavidin-HRP (SA-HRP) is introduced. Another washing step is performed, and then a TMB color substrate is added and incubated in a dark environment at room temperature to initiate color development. The intensity of the color reaction is directly proportional to the concentration of CXCL13 present in the sample. Finally, a termination solution is added to stop the reaction, and the absorbance value is measured at a detection wavelength of 450 nm (with a correction wavelength of 570-630 nm) using a microplate analyzer.

Interleukin 1 (IL-1) is an extracellular peptide weighing 17kDa. It comprises two proteins, IL-1α and IL-1β. IL-1β is a protein precursor that is secreted by activated macrophages. It is converted into its activated form through hydrolysis of caspase1 (CASP1/ICE). The cytokine IL-1β plays a crucial role as a mediator of the inflammatory response, impacting diverse cellular activities such as cell proliferation, differentiation, and apoptosis. These IL-1 proteins are part of an important family derived from biologically activated monocytes. They play significant roles in inflammatory and immune responses. When IL-1β expression is elevated, it can lead to various autoinflammatory syndromes, particularly CAPS, which is a single-gene condition caused by mutations in the inflammasome receptor NLRP3, triggering IL-1β processing.
