The Runt DNA-binding domain is present in this nuclear protein, which belongs to the RUNX family of transcription factors. It plays a crucial role in osteoblastic differentiation and the formation of the skeletal structure. Additionally, it serves as a scaffold for regulatory factors and nucleic acids involved in the expression of genes related to the skeletal system. This protein has the ability to bind DNA either as a monomer or as part of a heterodimeric complex, with a higher affinity in the latter form. The N-terminal region of the encoded protein contains two areas where trinucleotide repeat expansions may occur. Mutations in this gene, including those in the aforementioned areas, have been linked to cleidocranial dysplasia (CCD), a disorder affecting bone development. Various isoforms of the protein are produced due to alternate promoters and splicing events.
