Our Mouse TLR4 antibody has been produced using rabbits that were immunized with a synthetic peptide conjugated with KLH. This peptide was taken from the N-terminal region of the mouse TLR4 protein, spanning amino acids 25 to 55. The resulting antibody is highly specific and can be used to detect the presence of TLR4 in mouse tissues and cells. Our production method ensures that the antibody has a high degree of similarity to the original mouse TLR4 protein, making it an excellent tool for research applications.

Stability & Storage

To ensure optimum preservation, refrigerate the substance at a constant temperature of 2-8°C for a period not exceeding two weeks. For extended storage, it's recommended to keep the sample at -20°C in small aliquots to prevent recurrent freeze-thaw cycles.

Storage buffer

The provided polyclonal antibody is purified and supplied in PBS with a concentration of 0.09% (W/V) sodium azide. To achieve purification, the antibody is passed through a protein G column and subsequently eluted using high and low pH buffers. Immediate neutralization is then carried out, followed by dialysis against PBS to remove any impurities.

Target

Background

The innate immune response to bacterial lipopolysaccharide (LPS) is mediated by cooperation between LY96, CD14, and this particular protein. By partnering with MYD88, TIRAP, and TRAF6, it triggers NF-kappa-B activation, cytokine secretion, and the initiation of inflammation. Moreover, it plays a role in LPS-independent inflammatory reactions triggered by free fatty acids, specifically palmitate. Working together with TLR6, it promotes inflammation in monocytes/macrophages in response to oxidized low-density lipoprotein (oxLDL) or amyloid-beta 42, without the direct involvement of LPS. In this scenario, oxLDL or amyloid-beta 42 binds to CD36, leading to the formation of a TLR4-TLR6 heterodimer. Subsequently, this heterodimer is rapidly internalized and elicits an inflammatory response, resulting in the production of cytokines like CXCL1, CXCL2, CCL9 (via the MYD88 signaling pathway), and CCL5 (via the TICAM1 signaling pathway). Additionally, this process leads to the secretion of IL1B.
