Overview

Description

The iNOS enzyme plays a crucial role in producing nitric oxide (NO), which serves as a messenger molecule with a multitude of functions throughout the body. Within macrophages, NO works to promote tumoricidal and bactericidal actions. As a member of the NOS family, iNOS is actually induced by endotoxins and cytokines. Furthermore, the enzyme is stimulated by interferon gamma, which collaborates with lipopolysaccharide, tumor necrosis factor alpha, or interleukin-1 beta to increase its activity. iNOS functions as a homodimer, with a binding ability to SLC9A3R1. Notably, there are two isoforms of this protein generated by alternative splicing in human cells.

The protein known as HEP-NOS, or hepatocyte NOS, also referred to as HEPNOS, is an inducible enzyme called inducible nitric oxide synthase (iNOS) or inducible NOS. It is also known as MAC NOS or macrophage NOS. Its other names include nitric oxide synthase 2 inducible (NOS2) and nitric oxide synthase 2A (inducible, hepatocytes). This enzyme plays a crucial role in the production of nitric oxide (NO) in various cell types, including hepatocytes and macrophages. It is encoded by the NOS2 gene, also known as nos2 or NOS2_HUMAN. Additionally, HEP-NOS is associated with the activity of peptidyl-cysteine S-nitrosylase NOS2.

As component of the iNOS-S100A8/9 transnitrosylase complex involved in the selective inflammatory stimulus-dependent S-nitrosylation of GAPDH on 'Cys-247' implicated in regulation of the GAIT complex activity and probably multiple targets including ANXA5, EZR, MSN and VIM (PubMed:25417112). Involved in inflammation, enhances the synthesis of proinflammatory mediators such as IL6 and IL8 (PubMed:19688109).
