It consists of three main structural domains: an amino-terminal head, a proline-rich region, and a carboxy-terminal tail (1). In its inactive state, the head and tail domains of vinculin interact to form a closed confirmation. However, when vinculin is phosphorylated at specific sites such as Ser1033, Ser1045, Tyr100, and Tyr1065 by PKC-α and Src kinases, and when phospholipids bind to the tail domain, this interaction weakens (5,6). |
