Description | TLR family members, named after the closely related Toll receptor in Drosophila, play a crucial role in the innate immune response (1-3). These receptors recognize conserved motifs present in different pathogens and are responsible for initiating defense responses. Upon activation, the TLR pathway activates NF-κB, which in turn regulates genes involved in immune and inflammatory processes. Interestingly, TLRs and IL-1 receptor family members share a conserved region called the TIR domain, which spans about 200 amino acids. This domain plays a fundamental role in signaling. In addition, TLRs form associations with several cytoplasmic adaptor proteins that possess TIR domains, such as MyD88 (myeloid differentiation factor), MAL/TIRAP (MyD88-adaptor-like/TIR-associated protein), TRIF (Toll-receptor-associated activator of interferon), and TRAM (Toll-receptor-associated molecule). These associations lead to the recruitment and activation of IRAK1 and IRAK4. Together with TRAF6, IRAK1 and IRAK4 activate TAK1 and IKK, which subsequently triggers IKK-mediated degradation of IκB. Normally, IκB sequesters NF-κB in the cytoplasm to maintain its inactivity. This cascade of events ultimately results in the activation of NF-κB and the expression of immune and inflammatory genes. |
| Other names | Proline-rich vinculin and TIR domain-containing protein B, also known as MyD88 3, is a putative NF-kappa-B-activating protein 502H. It is also referred to as PRVTIRB or TCAM1_HUMAN. This protein, encoded by the Ticam1 gene, is a TIR domain-containing adapter molecule 1. It is involved in the induction of IFN-beta. Additionally, it is a toll-like receptor adaptor molecule 1 and plays a role in the Toll-interleukin-1 receptor domain-containing adapter protein-inducing interferon beta pathway. TRIF, another name for this protein, is a crucial player in the innate immune response. |

| Specificity | The total TICAM1 levels can be detected using the TICAM1 Antibody, which specifically targets the endogenous TICAM1 protein. By utilizing this antibody, researchers can accurately measure the amount of total TICAM1 present in a sample. The detection of endogenous TICAM1 levels is crucial for understanding its role in various biological processes. To determine the protein's function and its involvement in signaling pathways, it is essential to analyze the total TICAM1 levels using the TICAM1 Antibody. |

Background | Involved in innate immunity against invading pathogens. Adapter used by TLR3 and TLR4 (through TICAM2) to mediate NF-kappa-B and interferon-regulatory factor (IRF) activation, and to induce apoptosis. Ligand binding to these receptors results in TRIF recruitment through its TIR domain. Distinct protein-interaction motifs allow recruitment of the effector proteins TBK1, TRAF6 and RIPK1, which in turn, lead to the activation of transcription factors IRF3 and IRF7, NF-kappa-B and FADD respectively. |
| Tissue specificity | Ubiquitously expressed but with higher levels in liver. |
| Posttranslational modification | Phosphorylated by TBK1.Polyubiquitinated by TRIM38 with 'Lys-48'-linked chains, leading to proteasomal degradation. |
| Celluar localization | Cytosol;Endosome; |
