Overview

Description

During mitosis, SGO1 has a key role in maintaining the cohesion of chromosomes. It accomplishes this by preventing the premature dissociation of the cohesin complex from centromeres following prophase. At this stage, a significant portion of the cohesin complex dissociates from the chromosome arms. SGO1 potentially achieves this by inhibiting the phosphorylation of the STAG2 subunit of the cohesin complex specifically at the centromere region. This ensures that cohesin remains persistent at the centromere until it is cleaved by ESPL1/separase during anaphase.

Other names

Serologically defined breast cancer antigen NY BR 85, also known as SGO1 or SGO L1, is a protein that plays a crucial role in breast cancer progression. It has been extensively studied for its potential as a diagnostic marker and therapeutic target.
The discovery of NY BR 85 has opened new avenues for understanding the underlying mechanisms of breast cancer development. Researchers have found that this antigen is significantly elevated in breast cancer patients, suggesting its potential as a biomarker for early detection and prognosis.
In addition to its serological importance, NY BR 85 is also a key player in chromosome segregation and maintenance of genomic stability. It functions as a shugoshin protein, ensuring proper chromosome alignment during cell division and preventing premature separation of sister chromatids. Dysfunction of SGO1 has been associated with chromosomal instability and an increased risk of developing cancer.
Interestingly, there are multiple isoforms of SGO1, such as SGO1AB, SGO1CD, SGO1EF, SGO1GH, SGO1KL, and Shugoshin-like 1. Each isoform is thought to have distinct functions and may contribute differently to breast cancer progression.
Further research is needed to fully elucidate the role of NY BR 85 and its isoforms in breast cancer. However, the initial findings hold promise for the development of targeted therapies and improved diagnostic methods, bringing us closer to a future where breast cancer is effectively managed and treated.

Target

Background

During mitosis, a crucial process for cell division, there is a key player known as SGO1 (Shugoshin 1) that plays a central role in maintaining chromosome cohesion. This vital function is achieved by preventing the premature dissociation of the cohesin complex from centromeres after the prophase stage. It is important to note that during prophase, the majority of the cohesin complex dissociates from the chromosome arms. SGO1 ensures that cohesion is maintained at the centromere by inhibiting the phosphorylation of the STAG2 subunit of the cohesin complex.
By preventing the premature dissociation of cohesin, SGO1 ensures the persistence of this complex at the centromere until it is cleaved by ESPL1 (separase) during anaphase. This process is essential for proper chromosome segregation during mitosis, and its successful execution relies on the interaction between SGO1 and PPP2R1A.
Interestingly, the phosphorylation of SGO1 is necessary for various functions. Firstly, it is vital for the proper congression of chromosomes, which refers to their alignment at the center of the cell. Additionally, phosphorylated SGO1 plays a crucial role in the correct attachment of spindle microtubules to the kinetochore, a specialized structure on the chromosome that helps in its proper segregation.
SGO1 also contributes to the kinetochore localization of PLK1 (Polo-like kinase 1) and CENPF (Centromere protein F). Furthermore, it may be involved in the tension sensing mechanism of the spindle-assembly checkpoint by regulating the affinity of PLK1 for the kinetochore.
One of the isoforms of SGO1, isoform 3, is specifically involved in maintaining centriole cohesion, which is essential for controlling the integrity of the spindle pole.
Additionally, SGO1 is involved in the centromeric enrichment of AUKRB (Aurora kinase B) during prometaphase, a stage of mitosis preceding metaphase.
Overall, SGO1 is a crucial protein that orchestrates several processes during mitosis, ensuring proper chromosome segregation and maintaining spindle pole integrity. Its various functions rely on interactions with other proteins and its phosphorylation status.
