Overview | |
Description | The AP-2 alpha protein is a sequence-specific DNA-binding molecule that plays a crucial role in regulating gene expression. It interacts with inducible viral and cellular enhancer elements to activate or suppress transcription of selected genes involved in a wide range of biological functions including proper development of the eye, face, body wall, limb and neural tube. Specifically, AP-2-alpha is essential for early morphogenesis of the lens vesicle and activates the PITX2 P1 promoter transcription through association with the CITED2 coactivator to the PITX2 P1 promoter region. This factor binds to a consensus sequence and suppresses the expression of genes such as MCAM/MUC18, C/EBP alpha and MYC. Its importance is highlighted by the fact that the proper regulation of gene expression is essential for normal development and function of cells, tissues and organs. |

Application | The recommended dilutions for the various applications are as follows: for western blotting, dilute the antibody at 1:500 to 1:2000; for immunohistochemistry, dilute at 1:50 to 1:200; for immunofluorescence or immunocytochemistry, dilute at 1:100 to 1:500; and for ELISA using a peptide, dilute at 1:20000 to 1:40000. These dilutions have been optimized for optimal sensitivity and specificity of the antibody in each respective application. It is important to note that the dilutions may need to be varied depending on the sample type and experimental conditions. However, using these dilutions as a starting point will ensure accurate and reliable results. |

Target | |
Background | Sequence-specific DNA-binding protein that interacts with inducible viral and cellular enhancer elements to regulate transcription of selected genes. AP-2 factors bind to the consensus sequence 5'-GCCNNNGGC-3' and activate genes involved in a large spectrum of important biological functions including proper eye, face, body wall, limb and neural tube development. They also suppress a number of genes including MCAM/MUC18, C/EBP alpha and MYC. AP-2-alpha is the only AP-2 protein required for early morphogenesis of the lens vesicle. Together with the CITED2 coactivator, stimulates the PITX2 P1 promoter transcription activation. Associates with chromatin to the PITX2 P1 promoter region. |
Posttranslational modification | Sumoylated on Lys-10; which inhibits transcriptional activity. |
Celluar localization | Cytoskeleton;Cytosol;Golgi apparatus;Nucleus; |
UniPort | P05549/Q92481 |
